چکیده
Background. Long-term extensive use of glucocorticoids will lead to hormonal necrosis the femoral head, and osteoblasts play an important role in prevention osteonecrosis. However, there is no complete cure for head. Mesenchymal stem cell- (MSCs-) derived exosomes are widely used repair various tissue lesions. Therefore, aim this study was investigate mechanism dexamethasone- (DEX-) induced osteoblast apoptosis therapeutic effect human umbilical cord MSC- (hucMSC-) exosome mimetic vesicles (EMVs) on osteoblast-induced by DEX. Methods. The viability primary MC3T3-E1 cells were determined Cell Counting Kit-8 (CCK-8), FITC-Annexin V/PI staining immunoblot. intracellular levels reactive oxygen species (ROS) after DEX treatment measured 2′, 7′ -dichlorodihydrofluorescein diacetate (DCFH-DA) staining. In study, hucMSC-EMVs N-acetyl-l-cysteine (NAC) as measures. expression B-cell lymphoma 2-associated X, Bcl 2, HO-1, nuclear factor erythroid-derived 2-like 2 MAPK- signaling pathway osteogenic cell treated with Dex analyzed immunoblotting. Results. significantly ROS accumulation. MAPK-signaling activated treatment. intervention downregulated DEX-induced activation addition, can reduce Conclusions. Our confirmed that regulates inhibit apoptosis.

Therapeutic potential of genistein in ovariectomy-induced pancreatic injury in diabetic rats: The regulation of MAPK pathway and apoptosis
Objective(s): Genistein, as a phytoestrogen found in legumes, has several biological activities in general and anti-diabetic activity particularly. In this study, we investigated the effect of genistein on proteins involved in β-cell proliferation, survival and apoptosis to further reveal its anti-diabetic potential in the ovariectomized diabetic rat. Materials and Methods: We used three-month-...
متن کاملIsolation and Characterization of Human Induced Pluripotent Stem Cells-Derived Mesenchymal Progenitors
Purpose: Isolating human induced pluripotent stem cells (hiPS)-derived mesenchymal progenitors as a new source of mesenchymal cells which can differentiate into different lineages like adipose and bone. Materials and Methods: After 7 days of hiPS1 culture on matrigle coated dishes, spindle like cells around colonies were removed by cell scraper. These cells that had mesenchymal like morphology ...
متن کاملComparison of Characteristics of Human Amniotic Membrane and Human Adipose Tissue Derived Mesenchymal Stem Cells
BACKGROUND Mesenchymal stem cells (MSCs) are ideal candidates for treatment of diseases. Amniotic membranes are an inexpensive source of MSCs (AM-MSC) without any donor site morbidity in cell therapy. Adipose tissue derived stem cells (ASCs) are also suitable cells for cell therapy. There is discrepancy in CD271 expression among MSCs from different sources. In this study, the characteristics...
متن کاملThe effect of human wharton’s jelly-derived mesenchymal stem cells on MC4R, NPY, and LEPR gene expression levels in rats with streptozotocin-induced diabetes
Objective(s): Type 1 diabetes (T1D) is an autoimmune disease resulting from inflammatory destruction of islets β-cells. Nowadays, progress in cell therapy, especially mesenchymal stem cells (MSCs) proposes numerous potential remedies for T1D. We aimed to investigate the combination therapeutic effect of these cells with insulin and metformin on neuropeptide Y, melanoco...
متن کاملHuman Mesenchymal Stem Cells Derived from Adiopose Tissue and Placenta and the Adipocytic and Osteocytic Differentiation
Introduction: Mesenchymal stem cells can be isolated from adult tissues, such as the adipose tissue, or other sources. Among all these sources, adipose tissue because of easy access, and placenta due to its immunomodulatory properties, in addition to another useful properties, were attracted more attention to themselves. Isolation and comparing these two different sources can help us for acces...
